I drive up from the D.C. area and often exit at Cranbury (Exit 8A) to get the best Buffalo Chicken Cheesesteak at Original Steak & Hogie. This place is especially convenient if you are jumping off the Tpk to get to I-95 around Trenton towards PA.

The Parkway is a better alternative to the Tpk IMO... You can take it to the NY Thruway and over the TZ Bridge, rather than go through the GW Bridge and the nightmare-ish Cross Bronx Expwy. You can pick up the Parkway at the Tpk Exit 11.
If you take the Parkway, right off exit 161 is Bucu Burger, heralded as the best burger around. They get their meat from one of the top suppliers in the NY Metro, and the result is fabulous taste.

re: equal_Mark
Get off the turnpike at exit 11 as menton1 said. By going TZ, it also is easier to take the northern route through CT (Hartford instead of New Haven).
It's been a long time since I've done the drive, but if you want to stop in MA, there is plenty of stuff in Sturbridge, right about where you get on the MA Pike.

re: cantkick
you can get off at exit 9, take route 18 north a couple of miles to New Brunswick and stop in some places near Rutgers University for lunch (lots of places on Easton Avenue) If you're up for it check out the Grease Trucks on College Ave for a Fat Cat.
Or exit 9, take Route 18 north to Route 1 North and stop at Skylark Diner for takeout.
