You might also like going to the Main Street Market and eating at Orzo and perusing some of the great little shops. Not near Monitcello though, which is a bit out from Charlottesville.
Not too far away is the Belmont area. I have been to the Local there, which is good, but I don't believe is open for lunch, but not 100% sure.

if you can get in, the restaurant at the barboursville vineyards isn't far and is LOVELY.
the downtown mall is charming but i've personally had lousy luck with food there. admittedly i'm not a local - just visit a lot from richmond. so maybe i just order wrong. :
)i like maya in charlottesville. i know they do a brunch that's delicious (the vegetable potato cakes with smoked salmon are to die for) but i don't know about lunch. not far from the mall.
It depends what you mean by nice ambience and good food. Do you want elegant and upscale? Comfortable and accessible? Either way, if you only have time for one meal, be sure to go to the downtown (outdoor pedestrian) mall. It's the heart of the action, and very charming.

For casual outdoor seating, decent burgers or grilled ham and cheese, try Miller's (a Charlottesville institution) or The Nook. Miller's has an amazing friend green tomato salad. Sounds weird, tastes great.
For brunch with atmosphere try l'etoile for the higher brow (Sunday only) or Blue Moon Diner for the lower (Sat and Sun).
